The medium defined by Kao and Michayluk was designed to grow cells and protoplasts of Vicia hajastana at a very low population -density in liquid media. The inability of the plant cells to grow at a very low population density may be caused by excessive diffusion of metabolic intermediates into the medium. resulting in their dilution in the cell to a level below that required for survival. Vicia cells were able to grow at an initial population density of 1-2 cells/ml when the mineral salt medium was enriched with organic acids. sugars. sugar alcohols. growth regulators. amino acids and other organic compounds. The percentage of cell division could be increased by raising the -concentration of CaCl2 from 1 mM. as in Gamborg B5. to 5 mM. Calcium may play an important role in the proces of cell division because of its ability to preserve the structural and functional integrity of plant cell membranes.
